DMAE:
- The acute skin-firming effects of DMAE have been confirmed by quantitative measures of cutaneous tensile strength. Although its mechanisms of action in the skin remain to be elucidated, evidence suggests that the skin is an active site of acetylcholine synthesis, storage, secretion, metabolism, and receptivity. Muscarinic acetylcholine receptors have been localized to keratinocytes, melanocytes and dermal fibroblasts, whereas nicotinic acetylcholine receptors have been found in keratinocytes.
- DMAE is an analog of the B vitamin choline and is a precursor of acetylcholine, the firming action of DMAE is explained by the fact that its molecule, considered to be a precursor of acetylcholine, alters muscle contraction.
- The benefits of DMAE in dermatology include a potential anti-inflammatory effect and a documented increase in skin firmness with possible improvement in underlying facial muscle tone.

This peptide is a fragment of type I collagen molecule and is believed to serve as a biological increase degradation signal of the skin matrix. Indeed, when collagen is degraded, more of its small fragments get created in the body, including GHK fragment. When the key skin matrix-producing cells (fibroblasts) detect increased levels of GHK, they “assume” that the skin matrix is being lost at a higher rate and begin synthesizing it more vigorously. Thus, Pal-GHK is intended to stimulate skin matrix, leading to wrinkle reduction, skin firming and other aesthetic benefits.
- Another interesting point about this peptide fragment, is that it is a part of another well known skin care ingredient, the copper peptide Cu-GHK. Copper peptides are known to improve wound healing, activate skin remodeling, improve the structure of skin matrix, reduce scarring and exert other beneficial effects on the skin.
- Palmitoyl tetrapeptide consists of a short chain of four amino acids (a.k.a. GQPR peptide or glycine-glutamine-proline-arginine) connected to palmitic acid.
- Palmitoyl tetrapeptide is believed to work by reducing the production of interleukin-6 (IL-6) by the the key skin cells, keratinocytes and fibroblasts. IL-6 is a molecule that promotes inflammation, which, in turn, leads to faster degradation of the skin matrix and thus contributes to the development of wrinkles and loss of skin firmness and elasticity.
